摘要 间充质干细胞是一种具有自我更新能力的成体干细胞,广泛存在于骨髓、脂肪、外周血等组织中。间充质干细胞目前在临床上具有很多治疗作用,给多种疾病的治愈带来希望。最近的研究发现,间充质干细胞在治疗过程中通过释放一些小分子物质来发挥作用,其中外泌体携带着大量的生物活性因子在细胞间进行交流,并在生理和病理过程中发挥着作用。外泌体作为近年来无细胞组织工程的研究热点,可在一定程度上解决再生医学临床应用中遇到的问题,如种子细胞来源、数量及免疫排斥反应等,同时在疾病诊断以及治疗方面表现出极大的潜力。介绍外泌体提取、鉴定的方法及其在组织工程中的应用,并对其未来的发展进行讨论。

Abstract:Mesenchymal stem cells are adult stem cells with self-renewal ability, which are widely found in tissues such as bone marrow, adipose tissue, and peripheral blood. Mesenchymal stem cells have many therapeutic effects in the clinic currently, which brings hope for the cure of many diseases. Recent studies have shown that mesenchymal stem cells release many kinds of small molecular substances, among them, exosomes carrying large amounts of biologically active factors can mediate communication between cells and play a role in physiological and pathological processes. As a research hotspot of cell-free tissue engineering in recent years, exosomes have contributed to solutions in clinical applications of regenerative medicine to some extent, such as seed cell source, quantity and immune rejection, and at the same time in disease diagnosis and treatment. This paper introduces the methods of exosome extraction and identification and its potential application in tissue engineering and discusses its future development.
